Time course and localization of Foxp3+ Tregs in spleens and lymph nodes assessed by immunohistochemistry. We assessed the dynamic changes in the number of Foxp3+ Tregs in the spleens by immunohistochemistry. Tissue sections were stained using an anti-Foxp3 monoclonal antibody. The positive cells in the spleens were counted in 10 ocular grid areas within each section at a magnification of ×200, and total was divided by the number of ocular grid areas. We counted no less than 6 sections in one time point. We assessed the dynamic changes in the numbers of Foxp3+ Tregs in lymph nodes (LN) by immunohistochemistry. Tissue sections were stained using an anti-Foxp3 monoclonal antibody. The positive cells in spleens were counted in 5 ocular grid areas within each section at a magnification of ×200, and total was divided by the number of ocular grid areas. We counted no less than 6 sections in one time point.
