CYP treatment of AIT recipients increases the number of lymphocytes which produce IFN-γ in response to 4T1 tumor cells. A) ELIspot assays for IFN-γ producing cells from spleens of mice with established 10 day 4T1 flank tumors which were either untreated, or received AIT with 50 × 106 B/I activated lymphocytes, with or without CYP pre-treatment (100 mg/kg IP) on day 9. Splenocytes harvested from recipient mice 5 days after AIT were stimulated with 4T1 or MethA sarcoma cells and the number of cells producing IFN-γ were enumerated. Results are pooled from 3 mice in each group, mean values from paired wells are shown, and are representative of 3 replicate experiments B) ELIspot assays for IFN-γ producing cells from spleens of mice with established 10 day 4T1 flank tumors which were either untreated, received AIT with 50 × 106 B/I activated lymphocytes, with or without CYP pre-treatment (100 mg/kg IP) on day 9, immunized with irradiated 4T1 cells with or without CYP pre-treatment(100 mg/kg IP) on day 9. Splenocytes harvested from recipient mice 8 days after AIT were stimulated with 4T1 or MethA sarcoma cells and the number of cells producing IFN-γ were enumerated. Results are pooled from 3 mice in each group, mean values from paired wells are shown, and are representative of 3 replicate experiments.
