Retinoic acid does not affect Th2 cells differentiation in vitro. Naïve CD4⁺ T cells (CD4⁺ CD62L^hi CD25^-) were purified from DO11.10 mice by FACS. Isolated CD4⁺ CD62L⁺T cells were stimulated with OVA peptide _323–339 in Th2 skewing condition with/without different concentration ATRA (0.01-10 μmol/L) for 3 days. The cells were then stained with CD4 and IL-4 antibodies and analyzed by FACS. Dot-plot pattern showed intracellular cytokine staining. Data are representative of 3 independent experiments.
