RA CD11b⁺CD11c-Ly6C^{low/intermediate}are the suppressive population. BM cells were differentiated in the presence of GM-CSF with or without 100 nM RA for 7 days, stained with fluorescently-labeled antibodies against CD11c, CD11b⁺, and Ly-6C to determine relative expression of Ly6C on CD11b⁺ CD11c cells (A). Cells were then sorted based on relative Ly6C expression and purified populations co-cultured with antigen-specific T cell receptor transgenic T cells for 96 hours with media, peptide or LPS and fold change in proliferation shown for peptide-stimulated (B) or LPS-stimulated (C) co-cultures.
