  1. The percentage of CD4+ or CD8+ T cells that show a positive production of IFN-γ or TNF-α was determined by the number of cells above the anchor gate (cytokine producing) over the total cell number as described in Methods. The results are from 40 aged, 32 HIV+ and 48 normal donors; and show the mean ± S.D. The data obtained with the aged and HIV+-derived T cells represent experimental sets performed at different times with the same or different control donors (24 controls with each group). In all cases, PBMC were incubated for 15 hrs at 37° with PMA and anti-CD3 and either monensin or brefeldin as described in Methods prior to analysis by the flow cytometry. The p values were calculated with the Student's t test and refers to a statistical comparison of the aged or HIV+ CD8+ T cells and the control cells. 1Controls are young donors with no known medical conditions or diseases. No HIV+ or aged subject was part of the control group in this study.
