Y420 is required for Rlk tyrosine phosphorylation after TCR engagement. Jurkat-Tag cells were electroporated with either WT Rlk-GFP or Rlk-GFPY420F and twenty-four hours later, cells were stimulated with anti-CD3 antibody OKT3 for the indicated times. Cells were lysed and immunoprecipitated Rlk was immunoblotted with anti-phosphotyrosine (4G10) (top panel), or anti-Rlk (bottom panel).
