Effect of various peptides on refolding recombinant HLA-E. A Various specific peptides were tested for their ability to induce and stabilize the refolding of recombinant HLA-E. Peptides were added to the binding assay at 10 μM, the concentration required for maximal observed refolding for the known binding peptide, VMAPRTLVL (see Figure 1). The rat class I MHC molecule RT1-A1c was also used as a control to assess the validity of this refolding assay. Results shown are the average of 3 independent experiments. B HLA-E was refolded with 100 μM nonamer random peptide library and its induction ratio was determined and compared to two rat TAP-B associated classical Ia MHC molecules, RT1-A1c and RT1-Au. Results shown are the average of 3 independent experiments.
