Production of IFN-γ by human monocyte-derived DC DC were treated with CD40L (top left panel) or CD40L+IL-1β (top right panel) for 24 hours, monensin being present during the last 5 hours of incubation. After staining for surface markers and intracellular IFN-γ, DC were analyzed by flow cytometry. Large CD3- cells were analyzed (as shown in gate represented in bottom left dot-plot) to exclude T cells, and results show staining above isotype control (bottom right dot-plot), being expressed as percentages of CD1a+ cells expressing IFN-γ. Result representative of 4 experiments.
