CD81 costimulation occurs via direct action on T cells. PBMC were treated with magnetic beads for depletion of non-T cells, then T cells or non-T cells were treated for one hour on ice with 10 μg/ml 5A6 anti-CD81 mAb, washed twice, and re-mixed. The cultures were then stimulated for 6 h in the presence of 0.1 μg/ml SEB, and CD69 and IL-2 expression were quantitated by intracellular staining. Note that pre-treatment of T cells, but not non-T cells, resulted in augmentation of CD69 expression and IL-2 production. Error bars represent S.D. Results are representative of three experiments.
