Wnt3A inhibits in vitro B lymphopoiesis. BM CD133+CD10- HPC (A: assay 1) or CD10+ B progenitor cells (B: assay 2) were cocultured with a confluent layer of the murine stromal cell line MS-5 for 3 or 2 weeks, respectively, while treated with Wnt3A (100 ng/ml), Wnt3A + sFRP1 (2 μg/ml), Wnt3A + Dkk1 (500 ng/ml) or medium only. The number of resulting CD19+ B lineage cells in each sample was determined by quantitative flow cytometry. The percentage of CD34+ cells among the CD19+ cells were measured before and after culturing, with and without treatment with Wnt3A (C).
