Effect of human serum and delipidated human serum on the ability of bacterial-ligands or bacteria to induce TLR2 or TLR4 activity. HEK-TLR2-TLR6 cells or HEK-TLR4 cells were incubated with a dose range of FSL-1 or LPS respectively in the presence of human serum (HS) or delipidated human serum (HSdelip). FSL-1 (a) but not LPS (b) activity was inhibited by HS compared to HSdelip. c LPS was preincubated with HS or HSdelip for 8 h before addition of HEK-TLR4 cells. LPS-activity was significantly inhibited by HS compared to HSdelip. d HEK-TLR2-TLR6 cells were incubated with S. aureus, E. coli, S. thyphimurium, B. breve or L. salivarius in the presence of HS or HSdelip. HS significantly inhibited S. thyphimurium, B. breve or L. salivarius induced TLR2/6 activity. Each value represents mean ± SD of triplicates.
