ERAP1 deficiency causes enhanced inflammatory responses through an NF-κB independent mechanism. Bone marrow derived macrophages (5 × 105 cells) were plated into 24 well plates, and activated with 100 ng/mL LPS for various timepoints (15 min, 30 min, 1 h, 2 h, 4 h, 6 h). Cells were immediately fixed, permeabilized and stained for p-p65 using BD Phosphoflow Perm Buffer per manufacturer’s guidelines. p-P65 activation was assessed with Cytek Aurora followed by analysis in FlowJo (A).
