IFN gamma could potentially induce a decrease in perforin of T EM CD8 T cells. (A) HeLa 229 cervical epithelial cells and peripheral blood epithelial cells (PBL) co-culture set up. PBL were cultured alone in a transwell insert (left panel) or co-cultured with a monolayer of HeLa 229 cells (right panel) at the bottom of a well of a 12-well culture dish, in the presence or absence of 600 U/mL IFN gamma; (B) Gating strategy for CD8 TEM cell population from PBL. The CD8 TEM cells were identified by gating the cells based on FSC/SSC profile, followed by CD3, CD8 and CCR7 and CD45RA expressions; (C) Perforin expression of CD8 TEM cell subpopulation from PBLs co-cultured with IFN gamma-exposed HeLa 229 cells. Histogram overlay of perforin expression of control CD8 TEM cells derived from PBLs cultured alone in the presence (red) or absence (green) of IFN gamma is shown in the left panel and histogram overlay of CD8 TEM cells derived from PBL-HeLa 229 co-culture exposed (red) or not exposed (green) to IFN gamma is shown in the right.
